Would you prefer a long ass manga (like maybe about 30-40 chapters) or a shortish manga (say around 10 to 15 chapters)?
Like what the others say-- it completely depends on the plot. If it's good, I'll drown myself in any chapter you throw. If it isn't, I wouldn't even bother reading.

Would you prefer a story that's more serious or light (ie. lots of comedy)?
Mmm, you know Honey and Clover? It's the best example I have of something with a whole lot of comedy, and drama at the same time. Comedy is necessary; I'd think that would be a given. But I like the serious Emotion stirred now and then, hm?

Ohhgods, not another Fushigi Yuugi the-girl-eats-herself-to-death-and-isn't-that-pretty-but-everyone-loves-her-anyway line; just too annoyingly typical. How 'bout a triangle, within the essential characters? Or a square, if you can manage to pull it off? B.O.D.Y. is mostly about the couple (and Wild Ones), but see-- it gets boring...

Hints, lots of hints, as it progresses. I don't like the fun ruined too early, but right at the end would be close to idiotic.

Most importantly, do you prefer romance with another genre (like romance-action or romance-fantasy, romance-comedy etc. Basically stories like Vampire Knight, La Corda D'oro, and School Rumble) or a story that focuses MAINLY on romance with a bit of extra sub-plot that you don't really care about (Special A).
Romance, once again, must always have some comedy and drama. But I personally'd like that it ends there. your choice though?

Incredible-- I love twists. But twists never have to be tragic--just unexpected. Like say, another pairing (not the main expected one) works out in the end? Stuff like that are great. But then again, if you want to go all out with the boyfriend tripping off a bridge and landing head-first on a pile of jagged rocks right before the girlfriend's 18th birthday while her mother is dying, then go ahead. Just please for the love of all things sarcastic, do not leave the story hanging with only implications. That'd suck.

The main character is ALWAYS: very detailed, with accompanying very-detailed hair, strong, not-quiet, easy crier, easy blusher, dumb academically, and there is ALWAYS someone who is supposed to be prettier than her with long straight hair and slanty eyes. Unless smart and there to be loved by all intentionally. Geezus.

I'd say-- do whatever works for you. It's your main character and that means she should be your personal favorite. Just make 'er endearing and smart, please. I hate the guy being the cool-smart-no-one-can-beat-me one.

I'll answer based on the assumption you want to write/draw a shojo title, since I assume that's why you're asking. If not, my bad.

Blanket answer- Depends on the story and how it's written. But to be specific...

Well, depends on if you can write a good, long story. Some titles (see: Monster) are written so well you don't feel that one chapter out of its 162 chapters is out of place or makes the story too long. Other stories can be told in a chapter or two without feeling rushed or under-developed. What's important is that in your chapter span you have well-developed characters and an engaging story. There's no right page count to get that.

Depends on my mood. I like both.

I like romance at the middle- have enough building up of emotion to where they get together, but then follow the couple so you can see how their relationship develops.

A long ass manga XD It can be longer than 40 chapters, I don't care as long as the story is good.

I guess a light story that's like comedy/romance/action.... It's not like I dislike serious mangas but I don't like it when things are way to dramatic, like some drama queen bitching about everything and making a hen out of a feather.

Just romance between mainly two people. It's ok if there's some jealousy sometimes but I don't like it when I want the lead character to end up with a specific person and that person doesn't.

BEAUTIFUL! The rest is up to the manga-ka hahaha... I just can't stand ugly manga.... it takes alot of time to get used to reading something that looks horrible.

umm if you're talking about main girl character, she doesn't have to be extremely beautiful, just plain is good and a lot of books and mangas like to make the main girl character realy clumsy and stupid. i hate that, the main girl character should just be like an ordinary person.
the main guy character doesn't have to be really good looking, but he at least has to be good-looking (or else he will have no fangirls :P) his personality shouldn't be emo and cold like most guy characters in mangas. But his personality has to be opposite to the main girl character

If there's romance - between two people is better than most harem. This is only due to the fact that most harem I have read have more predictable plots. If this wasn't the case I really wouldn't care.

I like a story that has a great amount of focus on horror, mystery, or fantasy. Romance doesn't figure in so much as intrigue.
Ecchi - not necessary, but it can be a plus, as long as it's done right. (I suppose ecchi doesn't have much to do with romance for me XD)
Tragic endings are alright by me, although I'd rather be in the mood for one.
Don't care as long as she/he isn't too predictable in her/his pattern of behavior - I like a surprise. Even better, a full-fleshed character with a good narrative.

She doesn't have to be smart but not a total idiot either.
