I'd say Gurren Lagann. No other show has communicated the "believe in yourself and do the impossible" message quite as well as that show.

So far for 2012 , the most Inspiring anime would be Space Brothers.
With a lot of heart and luck and perseverance , one can reach ones goal. This anime makes me feel good inside too.
And the next one would be ,
Sakamichi no Apollon , even though its a josei title , I really got inspired to get back into music especially piano and drumming and learn how to play jazz..It reminded me how I love the music genre ,the art of jazz in its pure form of fun ,improvisation and supporting each other
