Why did you not include Baccano! in that list? DISAPPOINT.
Maria Holic wasn't that unique - It was just some yuri combined with some traps combined with SHAFT.
Incidentally, setting refers to the physical characteristics of the environment. E.g The setting of Seitokai no Yakuindomo is a bloody school.
The word you're after is premise.

Yojouhan Shinwa Taikei is another anime that hasn't been mentioned yet that's unique in it's plot direction and overall execution. Whilst the first eight episodes are engaging enough, it is at the climax of the series that all the thematic devices, plots, characters and storylines converge to give way to a rather satisfying and explosive ending. Whilst the traditional style of drawing may throw some people off, it's nothing people cannot handle if they've watched Kuchu Buranko. There's a reason most people on /a/ refer to it as the best anime of 2010. Actually on second thought, that's rather off-putting.
