Shoujo means that the protagonist is a teenage girl.
Shounen means that the protag is a teenage boy.
Josei means that it's aimed at girls. (Chihayafuru) Though that doesn't mean guys can't enjoy them, they tend to be better than all that ecchi shit that is currently airing.

Shoujo manga and anime is demographically targeted at girls aged 10-18, while Josei is aimed at an older female demographic (say 15+). Shounen and seinen have the same respective connotations except for male demographics.

Obviously there's a lot of reader/viewer crossover (I also really like Chihayafuru, for example), but that's the idea. This will usually mean female and male protagonists respectively, but I'm sure there are exceptions. Yaoi being a major one.

Also, bishounen and shonen-ai are pretty standard give-aways for a shoujo or josei.
