Power Armor/Suit ; This would be mecha a human sized individual would wear.
Fighter Class: This would be along the lines of a Gundam or Macross Veritech
Large Class: Larger than a fighter but smaller than an aircraft carrier. Think Mazinger, Gaiking, Voltron, etc.
Giant Class: Think the size of s large ship or two. Think Macross Quarter and the like.
Ridiculous Class: Think the size of a moon or larger. Unicron from Transformers, STTGL, Demonbane, etc.

Can't forget Super Deformed and model classes. (then we can talk small robots like the ones from robotics;notes or angelic layer that are controlled remotely...)

This is one of those questions that really depend on how you intend to use it and various other factors. The STTGL or Demonbane have it as far as sheer power. However, as far as effective use with crew and attachments, I would say the Macross Quarter from Macross Frontier. To be honest, we need to divide this up into classes. Something along the lines of:
Power Armor/Suit ; This would be mecha a human sized individual would wear.

I love that last classification. That sums it up perfectly and why STTGL is just absurd and lame!

Gurren Lagann is an auto win with me, as it's powered by awesome of course.

This would be a better question if you were asking who the greatest Pilot was, in which Amuro Ray takes the lead.

Answer: Play several games in the Super Robot Wars series and decide for yourself.

But I suspect the answer will end up being either Ideon or Zeorymer.

So far Demonbane and its incarnates are touted as the most powerful. There is also Getter Emperor, Ideon (who is a giant reset button), and Gurren Laganns final form in the top. There are also not too known units like many of the SRW series machines, which there are quite a few who use time and space as weapons.

Greatest pilot is Ozma Lee from Macross Frontier. Think of a new generation Roy Fokker who doesn't get killed.

I like Ozma, i really do, but he is no Roy Fokker :)

They really did write Ozma a lot like Roy, but I must say Roy was just slightly better
