I'll be in Tokyo for one night, arriving late afternoon, leaving the next morning.
Where should I stay? And what should I do?

Shinjuku Capsule hotel is cheap and great, it's a very different type of experience. Also, since you've got limited time, check out Akihabara while you can. You won't regret it. Lastly, if there's time, Shinjuku Gyoen is a beautiful park.

A single night... that's tough. A soapland works, if that's your thing. But, here's what I'd do assuming I'm near the Yamanote line:

1. Go to an all you can eat shabu-shabu joint first (take friends, or find some). There's a great place called Nabezo in Asakusa. There is a ridiculously awesome shrine nearby to check out before or after meal.
2. Punt around Shibuya for a bit. Check out the fashion, lights, general craziness. Many people will recommend Akihabara, but it closes early, so unless you get there before 8 pm, don't bother. Shibuya is a better bet.
3. Find a gritty izakaya (preferably on a lesser known stop on the Yamonote, like Yurakucho or Meguro), a place with good nihonshu and a cheap karaoke machine (preferably run by old people), and then pretty much just drink until everybody's your best friend. Eat yakitori and whatever else they recommend. Just say 'omakase' (bring me whatever you like).
4. Find a place to sleep (hotel or internet cafe).
5. Wake up. If already nearly afternoon, then find tonkotsu ramen (available at Narita airport, Kookai is the shit!). If you have time for 2 meals, eat convenience store food for breakfast (like onigiri or yakisoba sando), then find tonkotsu ramen (or alternately tsukemen or CoCoIchi Curry House).
6. Fly away. Wipe away single tear.
Addendum. Roppongi sucks.

Plan where you will stay before you arrive so you don't waste your time looking for a room.

+1 for convenience store breakfast.

Heck, I live off of train station bentos and convenience store food when I'm there.

Depends on if you will be in Tokyo 23 or on the whole other side of Tokyo-shi, or is it anywhere in the Tokyo prefecture? Akiruno is a pretty far ways away from Adachi but still in Tokyo-fu

Edit: Anyways, its mid summer, so goto one of those indoor water parks that will be packed or go see the imperial gardens, thats always nice. and both can be done in less than 3 hours each. for night you can (within the 23 wards) goto a club, the drinking age is 20 so it shouldn't be a problem, but even with its super low crime rate a 7 day club is where the trouble makers will be if you were to run into one

If I were in your shoes, I wouldn't even bother staying at a hotel or sleeping. One night? I'd go out and see the city and search the internet for what's open 24/7, that way you'll have something to do dead at night. But that's just me..

I agree. I would feel like I got ripped off of my time if I only had one day/night there. I would need to go live it up and try to find the craziest things possible to do.

You need to see the Robot Show.

It's bikini'd girls riding on top of robots that fight each other. It doesn't really get better than that.

Why for only one night?
