Sex is male, gender is female. Out of politeness & to avoid irritating others, I stopped using any gender pronouns at all for Grell because this usually initiated a political correctness war in the forums.

For PC reasons, some refer to one as their chosen gender. Others see it as fault in logic; If a puppy is raised by a cat & thinks it's a cat, do you refer to the puppy as a kitten? If Grell had a sex change operation, that would make a big difference, but they didn't have sex op surgery in the 1800s. Then again, we have seen 1960s computer technology in Germany in the most recent manga arc, so if Toboso is going to take that kind of liberty with tech time, who's to say Grell won't be a real woman someday? This series also does have magic.

Manga Grell is more violent, serious, is a minor character who hates Undertaker, likes William way more than Sebastian, when flirting there's more of an air of violent intent, there is also litlte to know crossdressing.

Anime Grell is a clumsy silly buttmonkey, a frequently reoccuring secondary character who falls in love with every sexy guy Grell comes in contact with, is overly romantic, & crossdresses sometimes.

You could say that they're almost 2 different Grells.
