The first five I can think of are things people have already said:

- Konosuba I enjoyed from the start, but I don't think the actual laughter started until the 'cabbage incident'.
- Teekyuu is one series that always bothers me how few people know of it. I think it really hit its high point around seasons 4 and 5, but I still distinctly remember how hysterically I found myself laughing after the uh... 'balls' comments in the first or second episode of season 1.
- Gekkan Shoujo Nozaki-kun is quite an obvious pick I guess, but it would be a crime to leave it off this list. The manga is just as laughter-inducing in my opinion, was kinda surprised by that (I expected it to be less funny because of the absence of the comedic timing and dry delivery etc.)
- Haven't You Heard? I'm Sakamoto was absolutely hilarious for the first few episodes, but unfortunately for me it sort of became less and less funny as it went on. There were still hilarious bits in the later episodes, but because it started spending whole episodes setting up for the jokes, it just wasn't as good as the more gag-comedy style earlier episodes.
- Tanaka-kun is Always Listless kinda had the opposite effect on me as Sakamoto; I started it just viewing it as a calm, cheerful, carefree Slice-of-Life show, but the further it went on, the funnier and funnier I found it.

As for others that haven't been mentioned:

- Working!! / Wagnaria!! is another series I feel doesn't get enough love. I am rediscovering through the new www. series how you have to get to know the characters for a few episodes before the jokes really start hitting their mark, but at it's high point, I think Working!! was just as funny, if not funnier in places, than Nozaki-kun...
- Prison School was a series that to be honest, I didn't really find all that funny at first. This is a bit weird to explain but sometimes with certain series I get this 'nervous' feeling that makes me kinda uncomfortable about certain situations (not the same as plot tension, I mean about specific potential developments - in the case of Prison School, it was about Kiyoshi going to meet Chiyo, and the whole uniform thing), but once it got past that bit of the plot, I started finding it more and more funny, and began to wonder more and more how I hadn't seen the comedy from the outset.

I know this is already unreasonably long so here are some other honourable mentions that I remember laughing at on multiple occasions:

One Punch Man
Kiss Him, Not Me
Monster Musume
Yuru Yuri

Bono Bono (the currently airing version)

If you've never seen Urusei Yatsura--as only us longtime keepers of the flame have --you don't know the historical foundations of anime comedy:

All time: Fullmetal Panic Fumoffu

This past year: Nanbaka, Haven't you heard? I'm Sakamoto

Special mention: Gintama, I find a lot of the jokes to be hit or miss, but when it's funny it's really funny.
