1) Country of origin.
2) Direction you read the panels.

Those are the only hard and fast rules. There are some tendencies, although there are obvious exceptions to each category:

-- A much higher percentage of American comics are super-hero focused.
-- A much higher percentage of Japanese manga are focused on the daily lives of (moderately) realistic individuals.
-- There are a much greater variety of manga aimed at an adult audience (hentai, seinen, josei).

Of course, there are manga about superheroes, comics about the daily lives and loves of characters (see: Archie), and comics aimed at adults (see: Walking Dead).

One is from japan and one isn't. That's how I separate the two.
