Considering "The Big Three" were all manga in shonen jump, doesn't it have to be a manga in shonen jump first?

Yup, pretty much. Other genres such as Seinen, Shojo etc. just don't compete in terms of mass popularity compared to Shonen magazines; especially as famed as Shonen Jump.

Right now, I really think Black Clover is a good contender. Will it sell hundreds of millions like One Piece or Naruto? I don't think so, but the fact that it's already selling over 100k in its first week for each new volume is incredible. Once the anime debuts later this year, it'll most likely see a significant boost in sales just like My Hero Academia for example.

I would say One Piece is still going strong...and now we got Boruto.

As for the third slot....probably My Hero Academia...

In my opinion, for a series to become part of "the big three", they have to be long-running ones like One Piece.
Maybe Boruto can take a spot there in the future.
