While starting out as a magazine model in 2001, So Yi promised herself that by the age of 30 she would still be participate in modeling only, then she would call it quits. But luckily So Yi got a break in television advertisements (commercial films (CFs) in South Korea), and her most popular commercial being for Samsung Anycall.

In 2004, Yoon So Yi auditioned for the action movie set in modern South Korea, Arahan. Arahan was based on a popular South Korean comic, and explored the consequences of unleashing an ancient evil spirit into a modern city. So Yi's character is part of a hidden school of ancient masters who train a naive young policeman to fight this evil force. She was not a martial artist and had to undergo strict training for four months in a film action school, where she injured her finger and ankle while training. The movie also involved a lot of wire work and elevated jumps, which was traumatic for Yoon who has a phobia of heights. Even though she was not a trained martial artist, Yoon So Yi is fond of sports and exercise. Yoon proudly recalls that she was an athlete in junior high, and could run the 100m dash in 16 seconds. So Yi is 5'6" and at one point weighed only 50kg. Yoon said she has always had a skinny body. She eats normally but in small portions, and loves traditional Korean food like kimchi, noodles and also has a weakness for pizza.

In 2005, So Yi played a beautiful young swords woman set in ancient China, the martial arts epic movie Shadowless Sword. When she's not doing action movies, Yoon So Yi is a star on Korean television dramas, such as City of Glass on SBS. So Yi has been appreciated for her roles in earlier television series like Goodbye Solo and Say You Love Me. Yoon So Yi says she's not much of a fan in watching dramas herself, and never goes to the theater. She's got a bubbly personality and doesn't like to cry, which is why she prefers curling up to watch mindless action flicks and tense thrillers. But Yoon did cry at the funeral of iconic Korean actress Choi Jin Sil, who committed suicide October 2, 2008, a tragedy which shook South Korea and neighboring nations where Choi was very popular.

Yoon So Yi is very active in charity and relief work like at the W Magazine's "Love Your W" gala event, held October 2007 to promote breast cancer awareness. JTS, a Seoul-based Buddhist international relief agency, is close to her heart. It uses donations to eradicate famine, disease and illiteracy through various projects and programs, around the world and So Yi herself is a Buddhist. In June 2008, the National Center for Missing Children co-sponsored a Green Ribbon for Hope Campaign. The participants gathered at the Cheonggye Plaza for the program, where Yoon and another actress Go Ara gave speeches and then joined in a walkathon. In August 2008, it was reported that Yoon So Yi's jaw was dislocated when her co-star Yoon Jung Ah accidentally hit her right cheek too hard during a scene. But after only two days So Yi was back shooting for her drama. So Yi shall replace Kim Min Jung's leading role as Joo Jae In for the 2009 comedic drama Hero.
